yes the engine is filled with coolant but when the water pump stops spinning it will no longer pump, so gravity will keep the coolant from coming back to the rad and the level will change. by coming back i mean fully circulating. So the reading should be read when the engine is at the same temperature otherwise there will be innacuracy just like when readin the dipstick.

In our cars the cooling system is always filled!!!
The reserve box next to the radiator is there to help the system only to overflow
If the car has somewhere leakage only in this case the engine sucks from reserve box.
Always the air bubbles are out of cooling system because of pump.
The fluctuation can happen if there are bubbles but the bubbles can be there when you just changed the coolant until works the radiator fan for couples time.

If there is no problem then coolant level never can be under the mark MIN.
When the engine is hot the coolant level might exceeds the mark MAX, it is normal because the hot liquids have the bent to expand and to overflow. It this case the expanded coolant overflows to reserve box.
